Aim: To prove that chlorophyll pigment is necessary for photosynthesis in the plants.
Requirements: A well-watered variegated leaf plant, beaker, water, bunsen burner, mentholated spirit or rectified spirit or ethanol, iodine solution, a white porcelain tile.
Theory: Photosynthesis is a physiological process of green plants which helps to prepare their food or carbohydrates. The experiment to prove that chlorophyll is necessary for photosynthesis involves an important step known as De-starching. De-starching is a step in which the starch present in the leaves of the plant will be removed by keeping the variegated leaves of a plant in the darkroom or closed cupboard for about 2-3 days.

Procedure:
1. A variegated ( the leaf that has both green and non-green parts) leaf plant is taken.
2. The place where green patches are present in the leaf, chlorophyll pigment is present while at the other colour patches present in the leaf contain other pigments but not chlorophyll.
3. Before starting the experiment to prove chlorophyll is necessary for photosynthesis, an important step has to be carried out is known as De-starching.
4. This step is done to ensure that previously synthesized starch in the leaves get consumed up because photosynthesis doesn't occur at that time as the plant is kept in the dark place.
5. After the de-starching step, the variegated leaf plant is kept in bright sunlight for about 6 hours.
6. After this, one of the variegated leaf is plucked out from the experimental plant.
7. Then Iodine test is conducted for this experimental leaf to find out whether the leaf prepared food through photosynthesis or not.

Observation: The green coloured leave will turn blue-black after iodine test showing that starch is being formed or photosynthesis is carried out. The yellow leaf does not turn blue-black, indicating that starch is not formed in or photosynthesis is not carried out. Only those patches which were earlier green in colour turn blue-black after starch test while non-green patches remain brown.

Result:
Presence of starch in green coloured patches shows photosynthesis has taken place and chlorophyll is necessary for it.
