Within any living organism, chemical reactions take place all the time, called metabolic reactions. Almost every metabolic reaction is controlled by enzymes. Enzymes are proteins that function as biological catalysts. Without enzymes, the reaction would take place very slowly, or not at all.
All enzymes work best at a particular temperature, each enzyme has a temperature range in which a maximal rate of reaction is achieved. This maximum is known as the temperature optimum of the enzyme. But they are made inactive by high temperatures because all enzymes are proteins that are damaged by heat. Different enzymes have different optimum temperatures. For most human enzymes, this begins to happen from about $40 °$ C upwards. As the temperature increases beyond this, the enzyme molecules start to lose their shape. The active site no longer fits perfectly with the substrate. It can no longer catalyze the reaction. The enzyme is said to be denatured.
Hence, for the above reason, the activity of enzymes was carried out at $40 °$ C.
